Genomic analyses in a rapid autopsy cohort study of patients with Melanoma identify the genetic and transcriptomic landscape of acquired resistance to MAPKi and immunotherapies, providing insights for the potential improvement of therapeutic strategies
mutated TCGA–SKCM cohort . We downloaded TCGA–SKCM CNA data from cBioPortal and compiled the frequencies of CNA genes . We applied Fisher’s exact test to individual-level amplified or deleted events, which were counted in a given RAM case if they were identified from at least one tumor of that case.
We performed Fisher’s exact test to compare the patient-level frequency of each SMG in the RAM versus TCGA–SKCM cohorts and used the FDR approach to adjust thevalues. We then performed GO enrichment analysis with the clusterProfiler package to detect the significant biological processes for expressed SMGs.
