Florence Pugh is set to join the cast for Dune: Part 2 and her presence in the film makes Dune 3 an even stronger possibility.

Rumors around Pugh's casting began in March 2022, and it's since been confirmed that she will be taking on the role of Princess Irulan, a key figure in Frank Herbert's Dune novels, in Dune: Part Two.
Director of 2021's Dune and Dune: Part 2, Denis Villeneuve has been open about his desire to make Dune 3. In an interview with Entertainment Weekly Villeneuve stated that he "always envisioned three movies" and expressed his desire to see Paul Atreides arc through to the end, which would require a third movie. While there's no confirmation of a third film in the series yet, Pugh's casting strongly suggests that plans for Dune 3 may be well underway.
While the character is integral to the book series - with pieces of her writing used as a framing device and making her the de facto narrator of Dune - Irulan doesn't appear in person until the very end of the novel. If Villeneuve is committed to faithfully completing his adaptation of the first novel only, then Pugh may not have much screen time in Dune: Part Two. This does, however, make Dune 3 a more likely prospect.
Princess Irulan takes a much more active role in the second Dune novel, Dune Messiah. By this point, she has become Paul's wife - though in name only as Chani remains his one true love - and also serves as one of his advisors. Irulan's role in Dune Messiah is far meatier, so Dune 3 is arguably more important for the character as well as the series as a whole, and would provide Pugh with a lot to sink her teeth into.
